Pharmaceutical Grade Water Systems: Purified Water and WFI Design, Validation and Supplier Selection

Purified water (PW) and water for injection (WFI) are the largest volume raw materials in most pharmaceutical plants. The system that produces them runs 24 hours a day and touches every batch. As an equipment manufacturer that has built and commissioned these loops, we know the questions that come up again and again: what train to use, what materials, how to validate, and how to compare suppliers without getting lost in the sales talk. This guide gives you the practical framework.

Purified water vs water for injection

PW is used in formulations, cleaning, and utilities where bacterial endotoxins are not controlled. WFI is used in parenteral products and has stricter limits on endotoxins and microbial counts. Both are produced from a feed water that meets drinking-water standards, and both run in a recirculation loop with a sanitary design.

  • Conductivity: USP/EP PW limit is 1.3 uS/cm at 25 C (after non-purgeable organics adjustment). WFI has the same conductivity limit but adds a 0.25 EU/mL endotoxin limit.

  • Microbial: PW typically below 100 CFU/mL in a cold loop; WFI below 10 CFU/100 mL.

  • TOC: both target under 500 ppb TOC, with a 50 ppb system suitability check.

  • Loop temperature: cold loops run 20-25 C, hot loops at 70-80 C to control biofilms.

The standard PW/WFI treatment train

A typical train is: raw water pretreatment, double-pass RO, EDI, then a storage tank and a recirculation distribution loop. For WFI, the common configurations are either a hot loop (storage at 80 C) or a cold loop with periodic thermal sanitization. Some plants still use distillation for WFI, but RO + EDI + a hot or sanitizable loop is now widely accepted when the feed water is suitable.

Storage and distribution loop design

The distribution loop is where most quality problems start. The rules we apply on every project are simple: keep the loop fully closed, maintain positive pressure, run turbulent flow (Reynolds number above 4000), avoid dead legs over 1.5 times the branch diameter, and slope the piping for full drainability. The storage tank uses a hydrophobic vent filter and is sized for the peak demand plus reserve, typically 1 to 2 hours of make-up capacity.

Materials and welding

PW and WFI loops are built in 316L stainless steel with a surface finish of 0.4 to 0.6 Ra micrometers on wetted surfaces. Orbital welding with a documented weld log and material certificates (EN 10204 3.1 or equivalent) is the standard. Elastomers are limited to validated materials such as EPDM or PTFE. When we quote a pharma loop, we include the full documentation set: material certificates, welding records, passivation certificates, and pressure test reports.

Validation and documentation

Buyers should expect a supplier to provide FAT (factory acceptance test) support, IQ/OQ documentation, and a commissioning protocol. During FAT we run the skid with water, record conductivity, TOC, flow, and pressure, and produce a witnessed test report. On site, we support IQ/OQ and provide the operating procedure, sanitization protocol, and preventive maintenance schedule. Ask your supplier for a sample documentation package before you sign.

Cost and lead time expectations

The price of a pharmaceutical water system is driven by the loop length, the number of use points, the materials, and the sanitization method. A hot loop with many distribution branches costs more to install and validate than a simple cold loop. Lead time for a factory-built skid is typically 6 to 12 weeks; the distribution piping is often the longest item because it is welded on site. Get a delivery schedule in writing and ask about spare-part availability.
